一 质粒简介:

1. 丝状真菌CRISPR/Cas9基因编辑
  可用于曲霉(如黑曲霉、构巢曲霉)等丝状真菌的基因敲除、定点突变或敲入,是目前曲霉基因编辑的常用工具载体。
2. 高效转化与无标记编辑
AMA1元件让质粒能在真菌中自主复制,无需整合到染色体,转化效率高;后续可通过FRT位点去除潮霉素抗性标记,得到无筛选标记的编辑菌株,便于后续多轮编辑。
3. 在大肠杆菌中进行载体构建
携带pBR322 ori和氨苄抗性,可在大肠杆菌中完成sgRNA载体的构建、扩增和测序验证,再转化到真菌中使用。
