质粒简介:

1. 构建p53基因敲除细胞模型
转染该质粒后,Cas9在sgRNA引导下切割p53基因的靶位点,细胞通过非同源末端连接(NHEJ)修复时会引入插入/缺失突变,导致p53基因移码失活,从而获得p53敲除的细胞系,用于肿瘤抑制机制研究。
2. 构建p53突变的动物模型
可直接用于小鼠胚胎的原核注射,快速构建p53突变的小鼠肿瘤模型,模拟p53缺失导致的肿瘤发生过程,广泛用于癌症研究。
3. 癌症机制与药物筛选研究
p53是关键的抑癌基因,该质粒可用于在细胞/动物中引入p53突变,研究p53功能丧失对细胞增殖、凋亡、肿瘤发生的影响,也可用于筛选靶向p53通路的抗癌药物。
4. CRISPR/Cas9技术的工具载体
