二:质粒简介:

1. RNA-based iPSC生成(无整合重编程)
传统病毒载体(如慢病毒)会将外源基因整合到宿主基因组,存在致瘤风险。而这个质粒的核心优势是:- 先通过T7启动子体外转录生成VEE复制子RNA;
- 转染体细胞后,RNA在细胞质内自主扩增,持续表达OSKM重编程因子;
- 完成重编程后,RNA会随细胞分裂逐渐丢失,无基因组整合,安全性更高。
2. 干细胞重编程研究
用于将成纤维细胞等体细胞诱导为诱导多能干细胞(iPSC),为疾病模型构建、再生医学、细胞治疗提供无整合的iPSC来源。
3. 多基因共表达工具
2A肽+IRES的设计,让4个重编程因子在同一转录本上按比例表达,保证重编程效率。
