一 质粒简介:

1. CRISPR基因编辑基础工具

常规的Cas9表达质粒,可配合gRNA(向导RNA),在各类哺乳动物细胞中完成基因敲除、基因敲入、位点突变等基因组编辑操作。

2.  CRISPR相关的RNA可视化与示踪 6个串联MS2茎环是这个质粒:- 转录后Cas9的mRNA上会携带MS2重复序列

- 与MS2-MCP荧光融合蛋白共表达,就可以在活细胞里实时追踪Cas9的mRNA定位、转运、翻译动态

- 也可用于追踪CRISPR转录本的亚细胞分布、降解动力学

3. 基因表达调控与高级CRISPR系统- 可配合dCas9(失活Cas9)变体,结合MS2-MCP效应蛋白,构建CRISPRa基因激活、CRISPRi基因抑制系统,精准调控内源基因转录

- 用于单碱基编辑、表观遗传修饰(甲基化、组蛋白修饰位点定点改写)的载体骨架

4. 共定位与多组分合成生物学研究

MS2-MCP系统可招募额外功能蛋白到Cas9所在的转录/基因组位点,实现多分子复合物的定点组装、细胞信号操控等合成生物学、分子细胞生物学机制研究。

5. 体外/体内真核细胞表达

依托CMV强启动子,可在几乎所有哺乳动物细胞系、甚至活体动物(体内转染)中高效表达Cas9,适配体外细胞实验、动物模型构建等不同场景。
