一  质粒简介:

1. Cas9蛋白表达

质粒携带由毕赤酵母内源组成型启动子  pPFK300  驱动的人源密码子优化Cas9基因,可在酵母中稳定表达Cas9核酸酶,用于切割目标基因组位点。

2. sgRNA克隆与表达

载体支持直接克隆含锤头核酶(HH)和HDV核酶的sgRNA PCR产物,核酶可在酵母中自动切割并释放成熟sgRNA,实现高效的靶向引导。

3. 游离型复制与无标记编辑

质粒含  CEN/ARS  序列,可在酵母中以游离质粒形式复制;编辑完成后,可通过无选择压力培养消除质粒,获得无外源抗性标记的编辑菌株。

4- 毕赤酵母的基因敲除、定点突变、基因敲入等基因组编辑操作

- 代谢工程改造(如优化蛋白表达、合成生物学通路构建)

- 无标记菌株构建,避免抗性标记对后续实验的干扰

- 适用于工业毕赤酵母菌株的改造,用于重组蛋白生产、天然产物合成等研究
