一质粒简介:

质粒主要用途包括:
 1. 高效外源基因表达
转染细胞后,T7转录生成的RNA可利用自身编码的nsp蛋白进行自我复制,在低剂量下实现比普通mRNA高得多的蛋白表达水平,GFP可作为表达效率的报告基因。
2. 诱导多能干细胞(iPSC)重编程
它是经典的无病毒重编程工具载体。通过替换EGFP为Oct4、Sox2、Klf4、c-Myc等重编程因子,可在体细胞中持续表达这些因子,高效诱导iPSC生成,且无需整合到基因组,安全性更高。
3. 自复制RNA疫苗研发的骨架载体
VEEV复制子是saRNA疫苗的主流骨架之一。可将EGFP替换为病毒抗原基因(如新冠、埃博拉、流感抗原),构建自复制RNA疫苗,实现低剂量、高免疫原性的抗原表达。
4. 细胞标记与示踪
自带的EGFP报告基因可用于:- 检测载体转染/转导效率
- 追踪活细胞中RNA复制和蛋白表达的动态过程
- 作为阳性对照,优化转染条件
5. 病毒复制子颗粒(VRP)包装
可通过共转辅助质粒,将复制子RNA包装成复制缺陷型病毒颗粒,实现高效的细胞转导,且颗粒无法复制,安全性高。
