1. 植物精准单碱基基因编辑

专门用于植物细胞的胞嘧啶碱基定向编辑,在靶位点高效引入C-T点突变,可用于:- 基因功能敲除(提前引入终止密码子)

- 内源基因定点氨基酸突变、蛋白功能改造

- 农作物ALS(乙酰乳酸合成酶)基因定向编辑创制抗除草剂突变体(质粒名称自带ALS靶点相关设计)

2. 遗传转化与稳定株构建

依托T-DNA骨架+潮霉素筛选,通过农杆菌侵染获得稳定整合碱基编辑器的转基因植物株系,可稳定遗传编辑性状。

3. 载体构建便捷性

Gateway重组系统+多克隆酶切位点(Asci、SpeI、PacI等),可快速替换不同gRNA,靶向植物的不同基因位点,批量构建编辑载体。

4. 物种适配

启动子、T-DNA、抗性组合,广泛适配水稻、拟南芥、玉米、小麦等多数双子叶/单子叶模式植物与农作物的基因编辑研究与育种应用。

- 区别于普通CRISPR-Cas9的基因敲除,碱基编辑器无需DSB(双链断裂),脱靶风险更低、编辑更精准,非常适合单氨基酸替换、精准单核苷酸多态性(SNP)创制。

- Hygromycin(潮霉素)相比卡那霉素等,对很多植物的筛选效率更高、背景假阳性更低。
