一 质粒简介:

来源于Lachnospiraceae细菌的Cas12a核酸酶,相比经典Cas9,更偏好识别T富集的PAM序列,可覆盖Cas9难以编辑的基因组区域,切割后产生粘性末端,编辑特性更特殊。

- 核定位信号(SV40 NLS、nucleoplasmin NLS):帮助LbCpf1蛋白进入植物细胞核发挥切割功能。

- SmR(链霉素/大观霉素抗性):大肠杆菌原核筛选抗性,用于质粒克隆、扩增与保种。

- attL1/attL2、attR5:Gateway重组克隆位点,可便捷地和不同启动子、gRNA表达盒、植物筛选标记载体进行位点特异性重组,快速组装定制化的多靶点编辑终载体。

- 原核元件(ori复制子、rrnB终止子、T7启动子):保证质粒在大肠杆菌中稳定复制、扩增,也支持T7体外转录。 

- 植物(单子叶+双子叶)精准基因敲除、基因敲入、大片段删减等基因组定点编辑。

- 多基因/多位点同时编辑:Cpf1自带RNA自加工能力,可一次载体搭载多个向导RNA,简化多重编辑实验。
