一 质粒简介:

1. 诱导型基因过表达

- pTA7001系统,搭载糖皮质激素(地塞米松Dexamethasone)化学诱导开关:- 无Dex诱导时,目的基因几乎本底不表达,本底泄漏极低;

- 施加外源Dex后,可快速、高效、可控地启动目标基因在植物体内大量转录表达。

- 可规避目的基因组成型过表达带来的致死、发育缺陷,实现时间可控的基因激活。

2. 克隆体系:Gateway高效重组克隆

- 载体骨架带有 attR1/attR2 + ccdB 致死筛选框,是Gateway目的载体(Destination Vector)

- 无需传统酶切连接,仅通过LR重组反应,即可快速将入门载体(Entry clone)上的目的基因精准、高通量重组装入该载体,大幅提升克隆效率。

3. 载体骨架与转化特性

- 具备T-DNA左右边界(LB/RB),可借助农杆菌,将T-DNA区的目的表达框稳定整合入拟南芥、烟草、水稻等多种植物基因组,获得稳定遗传的转基因株系。

- 同时带有原核复制子(oriV、oriT等),支持在大肠杆菌、农杆菌中稳定复制与接合转移。
