质粒简介:

1. shRNA 克隆:设计靶向目标基因的 shRNA 序列,通过 AgeI/EcoRI 酶切连接到 pLKO.1 puro 载体上。
2. 慢病毒包装:将重组 pLKO.1 puro 与包装质粒(如 psPAX2、pMD2.G)共转染 HEK293T 细胞,收获含 shRNA 的慢病毒颗粒。
3. 细胞感染与筛选:用病毒颗粒感染目标细胞,加入嘌呤霉素筛选,获得稳定敲低的细胞株。

4.pLKO.1 puro 是经过简化的版本(7050 bp),比经典 8.9 kb 的版本更短,可能是部分冗余序列被去除,核心的 shRNA 表达、慢病毒骨架和筛选标记功能完全一致(https://www.kelei-biology.com/product/15917.html)。
