一:质粒简介
1. miRNA靶标验证
将目标基因的3'UTR(含miRNA结合位点)克隆到hRluc下游,与miRNA共转染细胞。若miRNA能结合并抑制靶序列,海肾荧光素酶活性会显著降低,而萤火虫荧光素酶活性不受影响,通过两者的比值即可验证靶向关系。
2. RNAi效率评估
将siRNA RNA的靶序列克隆到载体中,转染细胞后检测海肾荧光素酶活性下降程度,快速评估RNA干扰的沉默效率,优化siRNA序列。
3. 基因表达调控研究
除了miRNA,也可用于研究其他调控元件(如RNA结合蛋白的结合位点、非编码RNA的调控序列)对基因表达的影响
4. 多克隆位点(MCS)
位于hRluc下游,包含NheI、EcoRV、PmeI、NotI等多个酶切位点,方便插入目标基因片段(如3'UTR、siRNA靶序列等)。
