基因激活筛选:可构建sgRNA文库,利用该质粒进行全基因组水平的基因激活筛选,发现调控特定表型(如药物抗性、细胞迁移)的关键基因。
 稳转细胞株构建:慢病毒载体可高效感染分裂/非分裂细胞,结合BSD抗性筛选,构建稳定表达dCas9-VP64的细胞系,用于长期的基因激活实验。
- 联合SAM系统使用:与MS2-p65-HSF1辅助质粒配合,组成SAM(Synergistic Activation Mediator)系统,进一步增强转录激活效率,实现更强的基因上调效果。
