一:质粒简介

大肠杆菌CRISPR-Cas9基因编辑设计的表达质粒,:
 1. 细菌CRISPR系统重建
与配套的pCRISPR质粒(携带自定义gRNA)配合使用,在大肠杆菌中重建功能性CRISPR-Cas9系统,实现靶向DNA切割。
2. 大肠杆菌基因敲除/修饰
通过Cas9对目标基因位点进行双链DNA断裂,结合宿主菌的同源重组或NHEJ修复,实现大肠杆菌基因组的定向敲除、突变或修饰,是细菌基因功能研究的常用工具。
3. 抗噬菌体/外源DNA防御
可在细菌中表达Cas9与靶向特定序列的gRNA,赋予宿主菌抵抗携带靶序列的噬菌体或质粒入侵的能力,用于菌株的生物防御改造。

二:CRISPR-Cas9实验流程
1:pCas9 质粒转化大肠杆菌
(1)冰上融化感受态细胞,加入 10-100 ng pCas9 质粒,轻弹混匀。
(2)冰浴 30 min → 42℃热激 90 s(电转则1.8 kV脉冲)→ 迅速放回冰上 2 min。
(3)加入无抗LB液体培养基,37℃摇床复苏 1 h。
(4)取菌液涂布 氯霉素抗性平板,37℃过夜培养。
(5)挑取单克隆,PCR验证质粒,得到含pCas9的稳定菌株。 
2:构建含靶标gRNA的pCRISPR质粒 
(1). 设计靶向目标基因的gRNA(需含NGG PAM序列,避免脱靶位点)。
(2). 将gRNA序列克隆到pCRISPR质粒的crRNA表达区(常用BsaI/ BsmBI酶切连接)。
(3). 转化大肠杆菌,用对应抗性筛选,测序验证正确的质粒。 
3:pCRISPR 质粒转化含pCas9的菌株 
(1). 制备含pCas9菌株的感受态细胞(或直接用新鲜单克隆制备电转感受态)。
(2). 加入 50-100 ng pCRISPR质粒(如需同源重组,可同时加入修复模板),重复转化流程。
(3). 涂布 双抗性平板(氯霉素 + 氨苄/卡那霉素),30℃或37℃培养过夜。
 4:阳性克隆验证 
(1) 挑取单克隆,接种双抗LB液体培养基,摇至对数期。
(2) 提取基因组DNA,用跨靶位点的引物做PCR扩增。
(3). 直接Sanger测序,或酶切验证(若修复模板引入了新的酶切位点),确认编辑成功。
