一质粒简介:

细菌基因无痕敲除/等位交换
 两次同源重组:
1. 第一次重组
将构建好的质粒(携带目标基因上下游同源臂)通过接合导入目标菌,利用质粒上的同源臂与细菌染色体发生单交换,使整个质粒整合到染色体上。用卡那霉素筛选,存活的菌株就是整合了质粒的菌株。
2. 第二次重组
将整合成功的菌株在无抗生素、含蔗糖的培养基中传代培养。质粒上的同源臂再次发生同源重组,将质粒骨架从染色体上切离。 如果重组发生在另一侧同源臂,目标基因会被删除,得到无痕敲除株;
如果重组发生在同侧,会恢复为野生型。利用  sacB  的蔗糖致死效应,可筛选出丢失了质粒的重组菌株,再通过 PCR 或测序验证基因型。
